【摘要】 目的观察玄参提取物对大鼠动脉硬化及其炎症相关细胞因子的影响。方法将大鼠随机分为正常组、模型组、玄参提取物低剂量组、玄参提取物高剂量组和布洛芬组。模型组及各给药组采用一次性腹腔注射维生素D3 60万单位/kg,同时每天灌服10 ml/kg脂肪乳的方法制备大鼠动脉硬化模型。玄参低剂量组和高剂量组分别每天灌服玄参提取物20.5 mg·kg-1·d-1和41.0 mg·kg-1.d-1,布洛芬组每天灌服布洛芬混悬液144 mg·kg-1·d-1,正常组和模型组每天灌服等量蒸馏水。给药70 d后处死大鼠,对血脂、TNF-α,IL-1β,IL-6,IL-10 及NF-кB进行测定,并观察动脉的病变情况。结果玄参提取物可明显降低模型大鼠胆固醇和低密度脂蛋白的水平,提高高密度脂蛋白与低密度脂蛋白的比值,降低炎症因子TNF-α,IL-1β,IL-6的浓度,提高抗炎因子IL-10的浓度,抑制动脉NF-кB过量表达,抑制模型大鼠动脉壁中膜的增厚。结论玄参提取物可通过抗炎和降血脂而发挥一定的抗动脉硬化的作用。

Abstract:ObjectiveTo study the effect of extract from Radix Scrophulariae (RS) against atherosclerosis and the relative mechanisms in rats.MethodsRats were randomly divided into 5 groups. They were the conrtol , model,low and high dose of RS extract and ibuprofen groups.The model rats were intraperitoneally injected with 600 000 IU·kg-1 vitamine D3 and intragastrically administrated high-fat emulsion 10 ml· kg-1·d-1 for 70 days.At the same time, the model rats treated with drugs were orally given 20.5mg·kg-1.d-1 ,41.0 mg·kg-1·d-1 RS extract and 144.0 mg·kg-1·d-1 ibuprofen suspension respectively.After 70days,the rats were killed, and the serum levels of TC,TG,LDL,HDL/LDL,TNF-α,IL-1β,IL-6,IL-10,the expression of NF-кB in rat aorta and the pathological change of aorta were measured.ResultsRS extract could lower the serum levels of TC,LDL-C, TNF-α,IL-1β and IL-6, increase the level of IL-10 and the ratio of HDL/LDL,inhibit the expression of NF-кB and reduce the thickness of the artery wall.ConclusionRS extract has anti-atherescle ratic effect.The mechanism may be related with inhibiting inflammation and lowering serum lipid.

随着国民经济迅速发展和人们生活水平的不断提高,卫生部1988~1996年疾病死亡统计资料显示,我国与动脉粥样硬化(AS)有关的疾病每年造成的死亡数占到总死亡数的35%~40%,是目前我国人们疾病第一位死因[1]。由于其发病机制尚未完全清楚,临床也无特效药物根治,因此,从中药中发掘抗AS的药物具有特别重要的意义。
玄参为玄参科多年生草本植物玄参Scrophularia ningpoensis Hemsl的根,是常用的传统中药。始载于《神农本草经》,被列为中品,性寒,味苦、甘、咸,可入肺、胃、肾经,具有凉血滋阴、泻火解毒之功效。用于热病伤阴、津伤便秘、目赤、咽痛等[2]。现代药理研究发现玄参在心血管系统具有广泛而重要的药理作用,而其抗动脉硬化的实验研究尚未见报道。
本实验旨在借助动脉硬化大鼠模型,观察玄参提取物对动脉硬化大鼠及其炎症相关细胞因子的影响,探索中药玄参提取物抗动脉硬化的作用及其机制。
