【摘要】 目的采用高效毛细管电泳(HPCE)法测定脑得生软胶囊的指纹图谱。方法运行缓冲液:30 mmol/L硼砂-50 mmol/L磷酸氢二钠,pH9.98;操作电压15 kV,检测波长260 nm,柱温25℃,采用压力进样,进样时间8 s。分析时间25 min。结果指纹图谱中标示了16个共有峰,其峰面积之和大于总峰面积的90%,方法精密度、稳定性、重复性均符合指纹图谱有关规定。采用夹角余弦法,计算10批脑得生软胶囊指纹图谱相似度结果均在0.93~1.00之间,但不同厂家生产的脑得生片有较大差别。结论该方法为脑得生制剂的质量控制提供了依据。

Abstract:ObjectiveTo develop the fingerprint of Naodesheng soft capsules by HPCE.MethodsThe background electrolyte consisted of 30 mmol/L sodium borate and 50 mmol/L sodium hydrogen phosphate and its pH was 9.98 ,applied voltage was 15kV,operating temperature was 25℃, the detection wavelength was at 260 nm and the analysis time was 25min.Results16 peaks were indicated.The peak area of these components was over 90% of total area in the fingerprint.RSD of precision and reproducibility was in the range of 5%. The results of similarity evaluated by cosine were 0.93~1.00 from 10 batches,but that of 3 batches of Naodesheng tablets from the other manufacturers were different .ConclusionThe method may provide reliable foundation of quality control of Naodesheng preparation.

脑得生软胶囊是根据脑得生丸(片)经工艺和剂型改革而来。脑得生丸(片)收载于《中国药典》2005年版Ⅰ部,由三七、川芎、红花、葛根、山楂等5味中药组成,具有活血化瘀、通经活络的功效。用于瘀血阻络所致的眩晕、中风,症见肢体不用、言语不利及头晕目眩;脑动脉硬化、缺血性脑中风及脑出血后遗症见上述证候者[1]。临床应用广泛,疗效确切。但其制备方法为传统生产工艺,质量可控性差、药物稳定性难以控制,因而对临床疗效和患者服用有较大的影响,不能适应现代临床用药要求。为此,本实验室对其活性部位进行了提取纯化[2],在此基础上研制成脑得生软胶囊。本论文采用高效毛细管电泳(HPCE)法对脑得生软胶囊进行了指纹图谱研究,并对不同厂家生产的脑得生片进行了指纹图谱分析比较,为脑得生制剂的质量控制提供科学依据。
1 仪器与试药
Beckman P/A system5000型毛细管电泳仪(美国Beckman公司);融熔石英毛细管柱75 μm×57 cm(河北永年瑞沣色谱器件有限公司);十万分之一天平(Sartorius BP211D)。 脑得生软胶囊(实验室自制,批号070801,070901,071001,071213,071214,071215,080815,080817,080824,080826);脑得生片,市售;葛根素购自中国药品生物制品检定,批号110752-200209;硼砂、磷酸氢二钠、氯化钠、氢氧化钠为分析纯,乙腈、甲醇为色谱纯,水为重蒸水。
2 方法与结果
2.1 电泳条件运行缓冲液:30 mmol/L硼砂-50 mmol/L磷酸氢二钠,pH 9.98;操作电压15 kV,极性由正到负,检测波长260 nm,柱温25℃,采用压力进样,进样时间8 s。毛细管冲洗方法:开机后用0.1 mol/L氢氧化钠溶液冲洗2 min,运行缓冲液3 min,每次测定之间运行缓冲液冲洗2 min。
2.2 对照品溶液的制备精密称定葛根素和芦丁对照品适量,用含0.5%氯化钠的60%乙腈溶液制成每毫升含50 μg的对照品溶液。
